MICROPROPAGACIÓN DE LAVANDA (LAVÁNDULA ANGUSTIFOLIA MILL.) A PARTIR DE YEMAS AXILARES MICROPOPAGATION OF LAVENDER (LAVÁNDULA ANGUSTIFOLIA) FROM AXILLARY BUDS. G.PORTILLA, J. B ; ELTRÁN y A. VEGA
RESUMEN Se describen los métodos para la propagación in vitro de lavanda (Lavándula angustifolia Mill). La micropropagación se realizó a partir de explantes de yemas axilares. La etapa de proliferación se llevó a cabo en el medio de MS suplementado con ANA, BA y combinaciones de éstos en presencia o ausencia de leche de coco y manteniendo un fotoperíodo de 16 h y una irradiación de 45 m E m² s-1. Los mejores resultados se obtuvieron con 1,15 o 3,15 mg L-1 de BA. Con esta última concentración hormonal, sin embargo, se formó una masa muy compacta de tallos con hojas pequeñas (inferiores a 0,5 cm); al subdividirla y transferirla a medio MS sin hormonas se obtuvo al cabo de 45 días un importante crecimiento de hojas y tallos lo que permitió la utilización del material para la siguiente etapa de enraizamiento. La inducción de raíces se logró en medios de MS con macronutrientes a la mitad de la concentración normal y 0,2 - 0,4 mg L-1 de AIB durante 7 - 14 días; posteriormente los brotes fueron transplantados a maceta y aclimatados en invernadero. Palabras claves Lavándula angustifolia, lavanda, micropropagación, enraizamiento in vitro. SUMMARY Methods for in vitro micropropagation of Lavándula angustifolia Mill are described. Micropropagation was performed from axillary buds. Propagation step was carried in MS medium supplemented with NAA, BA and combinations of them in the presence or absence of coconut water and mantaining the cultures under 16 h pholoperiod and irradiation of 45 m E m2s-1. The best results were obtained with 1.15 or 3.15 mg L-1 BA. However with the last hormonal concentration very compact shoots with small leaves were formed; when these mass of shoots were subdivided and transfered to a MS medium without hormones it was attained an important growth of leaves and stems after 45 days; then the vegetable material was utilized in the next rooting step. Root induction was obtained in MS medium with macronutrients at half concentration and 0.2-0.4 mg L-1 IBA during 7-14 days. Afterward the shoots were transfered into pots and aclimatized in a greenhouse. Key words: Lavándula angustifolia, Lavender, micropropagation, in vitro rooting. |
